qPCR slope to efficiency

With this online tool you can calculate the % of efficiency of the PCR reaction, the exponent and the amplicication from the slope produced by the quantitative PCR standard curve. Slope values between -3.1 and -3.6 are acceptable, and they will give reaction efficiencies between 90% and 110%. The formula to calculate the efficiency is …

Tm of long PCR primers

Calculate the Tm (Melting Temperature, in Celsius degrees) of your primers: For long primers use the formula :  Tm = 64.9 + 41(((G+C) -16.4)/N) Where N is the primer length in bases. This formula can also be used for shorter primers and is more accurate for long primers than the short primer formula.

ClustalW2

ClustalW2 is a multiple sequence alignment online tool provided by EMBL-EBI.

PlasMapper: plasmid drawing

Plas Mapper, a free plasmid drawing website, automatically generates plasmid maps providing the DNA sequenze (up to 20 Kb) in FASTA format.

Cloning: molar ratio of Insert to Vector

This online tool, provided by Promega, will allow you to calculate the molar ratio insert to vector, taking into account the lengh of the vector and insert (in kb) and the amount of vector (in ng).
